Photo-controlled calcium storm — Ca²⁺ overload in tumor cells

Photo-Controlled Calcium Overload from Endogenous Sources for Tumor Therapy

Record type
Preclinical scientific discovery (not a clinical trial)
Publication
Peer-reviewed
DOI
10.1002/anie.202317578
Editorial update
2026-10-29

Not a treatment recommendation. Experimental or ongoing research does not by itself replace standard care.

Scope / models
Multiple / solid tumors — Preklinik: tümör hücre hatları ve tümör implantlı fare modelleri
Molecular mechanism
Extracellular Ca²⁺ influx (TRPA1/ROS) plus ER Ca²⁺ release (NO → ryanodine receptor) cause cytoplasmic Ca²⁺ overload targeting mitochondrial failure. No exogenous Ca²⁺ source required.
Experimental models
Tumor cell cultures; injectable HA-coated mesoporous silica nanomodulator; tumor regression after NIR irradiation reported in tumor-bearing mice.
Translational distance
Not a human clinical trial or approved therapy. Selectivity, systemic toxicity, and translation into surgical oncology practice are unproven.

For patients

What is being studied? In the lab, a light-triggered method tried to kill cancer cells by flooding them with calcium. This is not a treatment tested in people.

Why promising? Using light to open endogenous Ca²⁺ channels and create an intracellular ‘calcium storm’ showed preclinical tumor-inhibition signals (Angew. Chem. Int. Ed. 2024; Ewha/China collaboration).

What is not proven? Human clinical efficacy, safe dosing, tissue selectivity, long-term toxicity, or superiority to standard care. This is not a treatment option.

Is this standard care today? No — this is a laboratory/scientific discovery dossier, not a treatment option.

For clinicians

DOI 10.1002/anie.202317578. Primary: photo-controlled endogenous Ca²⁺ overload. Press language uses ‘calcium storm’; no clinical claim. Physician: preclinical discovery card.

Limitations: Tek merkezli preklinik nanoteknoloji; insan verisi yok; cerrahi onkoloji endikasyonu yok.
