Not a treatment recommendation. Experimental or ongoing research does not by itself replace standard care.

For patients

What is being studied? Short immunotherapy before surgery may shrink some colon cancers. The parent trial still has open cohorts.

Why promising? Very high pathological response rates with neoadjuvant IO in locally advanced dMMR colon cancer.

What is not proven? Randomized phase III superiority; same strategy in pMMR; universal organ-preservation protocol.

Is this standard care today? This listing alone does not replace standard care; approval and guideline context require clinician judgment.

For clinicians

NICHE-2: n=111 efficacy; pCR 68%; major response 95%; NEJM 2024. Longer DFS/OS updates in Lancet stream — clinician verification.

Cancer / population: Colon — Metastazsız lokal ileri dMMR kolon

Intervention: Neoadjuvan nivo + ipi → cerrahi

Primary endpoint: Güvenlik (zamanında cerrahi) ve 3y DFS (NICHE-2)

Source-verified summary: Path response 98%; major 95%; pCR 68% (NEJM 2024).

Limitations: Tek kollu; ana NCT altında birden fazla kohort (karıştırma).
